Huyendo de los turcos, sus padres abandonaron la isla de Chío y fueron a Italia, acabando en Roma, donde tenían buenas relaciones (un tío suyo, Vicente, era general de los Dominicos, y en [[1570]] fue nombrado cardenal). Su padre era banquero.<br />
<br />
En [[1578]] Benedetto se doctoró en ambos derechos. En Roma era asesor financiero de Sixto V, que para agradecer sus servicios lo nombró cardenal en [[1586]]. Desempeñó diferentes cargos de gobierno en ciudades o regiones de los Estados Pontificios. En [[1611]] Paulo V lo nombró obispo de la diócesis suburbicaria de Palestrina. Como cardenal asistió a 7 conclaves.<br />

Cuando en [[1609]] falleció el Cardenal Torres, protector de los Escolapios, Paulo V lo nombró a él para sucederle, a petición de Calasanz y por sugerencia del P. Domingo Ruzola, carmelita, amigo de Calasanz y director espiritual del cardenal. Su papel fue fundamental durante los años que le quedaron de vida. Orientó la unión de las Escuelas Pías con la Congregación Luquesa, apoyó a Calasanz en la creación de la Congregación Paulina, recibió su profesión solemne en su mismo palacio en [[1618]], mandó al fundador a Narni a escribir las Constituciones, y finalmente recibió de nuevo la profesión de los votos solmenes de Calasanz ya en su lecho de muerte.<br />

Fue él quien pidió a Calasanz la fundación de la segunda casa fuera de Roma en Narni, feudo suyo, a sus expensas. Las escuelas se abrieron el 5 de noviembre de [[1618]]. A él le dedicó el P. Juan de Jesús María, Carmelita descalzo, el Liber de pia educatione, publicado en [[1613]], en agradecimiento por haber aceptado el cargo de protector de las Escuelas Pías.<br />
